Summary:

Hi everyone. I hope this post helps as I have tried really everything possible and have had great success in treating depression and anxiety. I always experienced severe anxiety, depression, and IBS my whole life (I am 28 right now). The issues were magnified significantly after taking Flagyl a year ago. Nothing helped my anxiety, depression, and IBS. Probiotics and extreme diets made it worse. The only thing that has helped so far has been the SD_ES donor. I have realized preparation is key to getting maximum benefits. More is less, and do not take more than two (000) capsules. Anything more caused brain frog.

The method that I have had incredible success:

You will get the whole frozen stool. You will have to take the frozen stool out with a sharp knife and cut it small enough to fit in capsules
Two delayed-release (00) frozen fmt capsules (This has caused heightened libido and overall GI help)

wait 10-20 mins

You will now take a cut stool piece and put it in a (000) capsule and let it rapidly thaw 5-10 in a baggy in warm to hot bowl.
One instant release (000) fmt capsule - you will rapidly thaw this frozen stool to a warm temp. Dont fill whole capsule - you only need about a half of capsule of stool (this is what CURED my depression).

The delayed release (frozen) was never able to cure my depression but you can try it first. I needed an instant release (thawed capsule).

One to two delayed-release (00)
I liked leaving off with a delayed-release capsule (frozen). I think the delayed-release frozen capsules crowd out some unwanted microbes that I noticed from thawed (slight bloating), but the thawed instant release was 100% needed for the extreme mental benefits.

Overall - delayed-release frozen capsules feel the best, but to get the maximum benefit, you need a thawed instant combo. Of the dozens of FMTs I did, I felt the most incredible benefit was doing a frozen delayed capsule ---> into a thawed instant-release capsule ---> wait two days ---> finish off one to two delayed-release capsules.

That sounds dangerous. The stools come split into 10 pieces. Defrost one piece before making capsules with it. If you have extra capsules from the 1 piece, re-freeze them since they'd probably melt in the refrigerator. An additional freeze-thaw cycle may decrease efficacy a bit but it's safer than trying to cut frozen stool with a knife.

I'm confused by this process. You're thawing the stool after you encapsulate it? There is no need to thaw capsules (or stool) prior to ingestion.

The delayed release (frozen) was never able to cure my depression but you can try it first. I needed an instant release (thawed capsule).
Frozen/thawed is not related to instant/delayed release. There are specific delayed-release (enteric-coated) capsules that are designed to not break down in the stomach, but rather further down in the small intestine.

I'm skeptical that thawing the stool first was what made the difference.

Ok, so based on this part, you tried both:
  • Delayed release capsules + fresh stool
  • Delayed release capsules + frozen stool
  • Quick release capsules + fresh stool
  • Quick release capsules + frozen stool

Frozen stool in a delayed-release capsule shouldn't be any different from fresh stool in a delayed-release capsule since the stool should defrost before the capsule breaks down, either way.
 
Of the other HMorg donors you tried (FL-RS, OR_JO, and UT-AW), how would you rank them relative to each other (It's pretty clear from this thread that all three rank below SD-ES-2015)?
 
Just in case it helps, I used a knife to slice frozen sample into the desired size, and it actually felt safe to me. However, the knife was more akin to a butter knife (not sharp), and the sample had antifreeze so that may have made it softer. I pushed on the back of the blade with my free hand and rocked the knife handle up and down to slowly and carefully push the blade through the sample. It was like working a knife through a piece of chocolate. This let me control the dose more easily. But obviously I would be very careful with anything than could potentially cut skin in this context, and if the sample was too hard then I wouldn't even bring a knife into the picture.

Why not just order them split into 10 pieces? In my experience, each piece was enough to make atleast 15 capsules (size 0) or for an enema. This has saved me so much time in the past.

Where did you get the stool from? Humanmicrobes.org donors split them into 10 pieces by default.

What surface did you put the stool on when cutting it? I have had a frozen solid stool (no antifreeze) on hand before and it was impossible to break up without defrosting it first. Much less with a butter knife.

The samples were all from UT-AW, and each were split into pieces, but less than 10 (at least that was the case for the first order—I don't remember the others as well). But the sizing of the pieces in the first sample was close enough to what you described in our email exchange, and keep in mind this was way back in early 2022, so I don't know if you changed your protocol since then.

I don't remember which, but I cut it on top of either a plate or a metal countertop while it was still inside the plastic bag it came in. I just remember being able to slowly drive the knife through the sample and I didn't even feel worried that it was going to cut through the bottom of the plastic bag underneath. Maybe it was the antifreeze that made it soft enough.

Glad you had so much success! Just wondering, how are you not sure your success is from a more compatible donor rather than thawed vs. unthawed? Also did the results stick?
