Hi, I will provide a bit of my backstory. I was born with severe health issues (Gut issues like food intolerances and SEVERE bloating after eating 99% of foods and especially fibre.

This worsened as I aged and I got diagnosed with SEVERE IBS-M leaning more towards constipation. And I also suffer from psychiatric issues and Chronic Severe Fatigue. Metabolic Issues and Hormonal Issues).

My maternal side— EVERYONE born through my Maternal Grandmother have similar health issues. Their kids have severe similar health issues but worsened + other additional health issues.

Now, I finally found a donor who lives near my area. He agreed mainly due to the payment I am providing.

Donor details/Characteristics—
• Model and Does Fitness Coaching
• Age— 33
• Height is 6'2" & decent bone structure. Very fit and muscular. Gym everyday and proper diet. Great hair and good skin.
• Sports history- Played soccer, baseball and powerlifting. But never been Competitive enough in sports. Never had the athletic drive but Always been active and fit though. No energy issues. That's it about his sports history.
• Always Been fit and on the lean side, just gained weight for his powerlifting phase (most of it was muscle) and no Metabolic issues or Energy issues.
• Have perfect beard growth for a man, indicating good hormonal levels.
• Great Hair Quality. Smooth Hair, Very dense hair and never had hair issues.
• Personality traits— Bit Cold Nature, Mix of Extrovert and Introvert. Calm and no mood issues at all.
• Sexual Activity— Doesn't engage in risky sexual activities, Hookups. He had One-Night Stands in past but is now in stable relationship. (Will test him for STDs too)
• Drug Usage— Alcohol once or twice every 2 weeks. Ciggerates occasionally. Nothing else
• Steroids/PEDs usage— Has only used Growth Hormone in the past for almost 2 years when he was around 25.
• Diet— Lots of Fruits, Veggies, Rice, Curd, Milk, Meat once a day, Not much spices and condiments and No gluten at all.
• No food intolerances apart from that He Avoids Gluten because it causes him increase in water-weight and overall Body and facial bloating.
• Antibiotic Usage— Has used antibiotics in the past but only for flu/fever and short term stomach poisoning.
• Family History— Siblings are healthy too and Mother is in good health accordingly for her age. Father has also been always healthy and fit.
• Important Thing about diet— He consumes 200-300 mg of caffeine because of hectic schedule.

~ MAIN OBSTACLE— His STOOL TYPE. I assumed that his stool would be type 3-4, firm and dark brown. But his stool is CONSISTENTLY TYPE 4, SEMI-FIRM/SOFTISH and DARK YELLOW.

Now, the only REASONABLE theory for his stool type could be the AMOUNT OF HIS CAFFEINE CONSUMPTION. His Hair is great, Skin is decent and his strength is good and decently muscular especially for his height.

So, I am not sure if I should use his stool. Because I have severe IBS and weak immune system. My mood and mental issues is very sensitive as in they can be affected by 99% of the food items available out there. Severe Chronic fatigue too.

Ideal and Healthy stool should be type 3-4, firm & dark brown. HIS STOOL CHARACTERISTICS (SOFTISH, TYPE 4 and CONSISTENTLY DARK YELLOW) indicates some kind of bile issues and missing microbes ? Or pathogen ? Or maybe the amount of caffeine He consumes daily ?

I have tried my best to explain everything in simple terms and have included almost all the information. Also, have formatted my post decently I suppose.

Doing all this while struggling with my severe health issues. So, I hope the post explains everything decently. I have tried my best.

Red flag. Many people drink caffeine and don’t have their stools affected. Food sensitivity is a sign of dysbiosis.

The fact that he felt the need to use HGH, drinks alcohol, smokes occasionally and others suggest he is low quality. And also needing caffeine.

You have CFS and multiple intolerances like Michael. You probably need a very high-quality donor like him.

This guy is not.

Only food sensitivity He mentioned is Gluten causing extra water weight and Overall Body bloating.

He says he needs to be lean and zero bloated for Modelling shows and gigs.

He said he will share his stool photos tomorrow. So, I can decide.

His Hair is really great, he is vascular and decent amount of muscles. Also good strength. His skin is decent too. His libido is high too.

You think Alcohol could be the issue or ciggerate ? But it's not daily though. Only weekly or every 2 weeks. Or maybe he is hiding his ciggerates use which could be of daily use.

Yes, I am in exactly the same boat/health status as Michael is. Probably worse in terms of mood and weight aspects.

I agree, Donor isn't as perfect as I think he is. Especially with all the Caffeine, Alcohol and Ciggerate usage. (The Ciggerates could even be daily use which he is hiding.)

Also, Is it allowed to share Donor Pics and Stool Pics here ? I think it would make this a lot easier.

I am not Michael, but I am 100% sure he wouldn’t allow that because that is private information. You can try contacting him privately through email though, but considering his current policy of not helping individuals unless they have a very good donor or are multi-millionaire investors, I doubt you would get much help.

I am sorry to burst your hopes, but its clear already that he is not an ideal donor for you. No need for pics if you can already tell from the questionnaire and stool info you provided.

You might want to get whole genome sequencing to check if it's a genetic issue. A consistent maternal inheritance pattern of chronic fatigue issues would suggest an mtDNA mutation.

Can that appear in an Ancestry.com result or is that expensive?

Interesting. Doubt CRISPR could fix that though, pretty sure other people with CFS may have tried that route before but gotten nowhere.

Ancestry.com is not WGS, it just tests common SNPs, so it would be useless for this purpose. It would cost around $500. I used Nucleus. Other companies are Mitohealth and Sequencing. Depending on the mutation, there may or may not be certain drugs and supplements that can work around the issue. At a minimum if there is a confirmed mtDNA pathogenic mutation it would tell him that his problems are not due to a gut microbiome problem.

Why would you say that when the gut microbiome is heritable, largely from the mother?

How is that possible when the gut microbiome regulates genetic expression and virtually every aspect of human health, development, and function? https://humanmicrobiome.info/genetics/

He does have gut issues that run in the family though. There is a confirmed gut microbiome issue.

But I get what you are saying. That the root cause is genetic and the gut microbiome is affected by the genetic mutation, instead of it being the root cause.
We can't say what the root cause is based on the evidence he gave; I'm just saying the root cause could be genetic.

I'm not ruling out a gut microbiome cause. I'm just saying genetics is a potential cause that should be checked

Disorders with a purely genetic cause are not just possible, but well-known and constitute basic scientific consensus. For example, Huntington's is caused by defects in the HTT gene. Full stop. No dysbiosis needed. Are you claiming otherwise? I said nothing about gene expression, rather, changes to the nucleotide sequence. Dysbiosis can surely cause a lot of disease, but to say it is the sole cause of all disease is not correct (I'm not sure if this is what you intend to imply here). Anyone trying to diagnosis a chronic health condition should examine all potential causes, including microbiome, genetics, nutrition, mitochondrial function, etc.
